O existenci DNA, jej\u00ed struktu\u0159e a \u00faloze se u\u010d\u00edme jako o faktech; uzn\u00e1vaj\u00ed je a schvaluj\u00ed v\u0161echna v\u011bdeck\u00e1 pracovi\u0161t\u011b. Ale co kdy\u017e v\u00e1m \u0159eknu, \u017ee DNA za\u010dala jako koncept. V roce 1869 Johannes Friedrich Miescher, l\u00e9ka\u0159 a biolog, jako prvn\u00ed v\u011bdec “izoloval” nukleovou kyselinu, kterou tehdy pojmenoval nuklein.<\/p>\n\n\n\n

Mieschera zaujala nov\u00e1, rozv\u00edjej\u00edc\u00ed se v\u011bda biochemie, v\u011bda, v n\u00ed\u017e se na biologickou hmotu aplikovaly chemick\u00e9 l\u00e1tky. Za biochemii reakce biologick\u00e9 hmoty na chemick\u00e9 l\u00e1tky, postupy a vznikl\u00e9 vedlej\u0161\u00ed produkty poskytovaly vod\u00edtka o slo\u017een\u00ed a struktu\u0159e bun\u011bk a jejich obsahu. Miescher v\u011b\u0159il, \u017ee bu\u0148ky v sob\u011b obsahuj\u00ed n\u011bco \u017eivotn\u011b d\u016fle\u017eit\u00e9ho, co se tak\u00e9 pod\u00edl\u00ed na procesu d\u011bdi\u010dnosti. Hoppe-Seyler, biochemik a majitel laborato\u0159e, navrhl Miescherovi, aby se p\u0159i sv\u00fdch pokusech zam\u011b\u0159il na leukocyty (b\u00edl\u00e9 krvinky). Miescher se \u0159\u00eddil Hoppe-Seylerov\u00fdm n\u00e1vrhem a odeb\u00edral leukocyty z hnisu na \u010derstv\u00fdch chirurgick\u00fdch obvazech z\u00edskan\u00fdch z nedalek\u00e9 kliniky.<\/p>\n\n\n\n

Miescher izoloval leukocyty namo\u010den\u00edm a promyt\u00edm obvaz\u016f v roztoku s\u00edranu sodn\u00e9ho a jejich p\u0159efiltrov\u00e1n\u00edm p\u0159es f\u00f3lii. Aby odstranil bun\u011b\u010dnou st\u011bnu a cytoplazmu, leukocyty n\u011bkolikr\u00e1t promyl roztokem kyseliny chlorovod\u00edkov\u00e9. J\u00e1dra odebran\u00e1 v p\u0159edchoz\u00edch kroc\u00edch energicky prot\u0159epal v roztoku \u00e9teru, aby odstranil p\u0159\u00edpadn\u00e9 zbytky cytoplazmy. K j\u00e1dr\u016fm z\u00edskan\u00fdm v p\u0159edchoz\u00ed f\u00e1zi byl p\u0159id\u00e1n uhli\u010ditan sodn\u00fd (alkaliz\u00e1tor) a pot\u00e9 kysel\u00fd roztok. Nuklein, DNA, byl pevnou \u010d\u00e1st\u00ed obsahu ve zkumavce, “sra\u017eeninou” v roztoku. Sra\u017eenina se tvo\u0159ila, kdy\u017e byla sou\u010d\u00e1st\u00ed roztoku kyselina, ale po p\u0159id\u00e1n\u00ed z\u00e1sady se rozpustila. Tato reakce, tedy tuhnut\u00ed l\u00e1tky p\u0159i okyselen\u00ed a rozpou\u0161t\u011bn\u00ed p\u0159i alkalizaci, nebyla nikdy p\u0159edt\u00edm pozorov\u00e1na.<\/p>\n\n\n\n

Aby prozkoumal slo\u017een\u00ed sra\u017eeniny, Miescher ji sp\u00e1lil. Na z\u00e1klad\u011b vedlej\u0161\u00edch produkt\u016f vznikl\u00fdch p\u0159i spalov\u00e1n\u00ed dosp\u011bl k z\u00e1v\u011bru, \u017ee nuklein obsahuje velk\u00e9 mno\u017estv\u00ed fosforu (ve form\u011b kyseliny fosfore\u010dn\u00e9) a dus\u00edku, nikoli v\u0161ak s\u00edry (s\u00edra se nach\u00e1z\u00ed a je v\u00e1z\u00e1na p\u0159edev\u0161\u00edm na b\u00edlkoviny). Na z\u00e1klad\u011b t\u011bchto zji\u0161t\u011bn\u00ed, ale tak\u00e9 na z\u00e1klad\u011b reakce sra\u017eeniny na kysel\u00e9 a z\u00e1sadit\u00e9 chemik\u00e1lie prohl\u00e1sil, \u017ee objevil novou l\u00e1tku, a uvedl, \u017ee “podle zn\u00e1m\u00fdch histochemick\u00fdch fakt\u016f jsem musel takov\u00fd materi\u00e1l p\u0159ipsat nukleinu”.<\/p>\n\n\n\n

Vydavatel \u010dasopisu, Hoppe-Seyler, zopakoval Miescherovy pokusy a potvrdil jeho zji\u0161t\u011bn\u00ed.<\/p>\n\n\n\n

Hoppe-Seyler z\u00edskal pro sv\u00e9 pokusy leukocyty z b\u0159icha ps\u016f. Psi byli na\u0159\u00edznuti v oblasti b\u0159icha a do t\u011bchto \u0159ez\u016f byly vlo\u017eeny \u010do\u010dky. Zbytek nasb\u00edran\u00e9 hmoty rozsekal, va\u0159il (ve vod\u011b a v alkoholu), okyseloval, alkalizoval, o\u0161et\u0159oval um\u011blou \u017ealude\u010dn\u00ed tekutinou, \u00e9terem a hork\u00fdm alkoholem a pozd\u011bji p\u00e1lil, aby mohl zkoumat a dokumentovat vedlej\u0161\u00ed produkty. Podle Hoppe-Seylera maj\u00ed kvasinkov\u00e9 bu\u0148ky podobnou strukturu jako bu\u0148ky hnisu. Kvasinkov\u00e9 bu\u0148ky podrobil podobn\u00fdm pokus\u016fm jako s ps\u00edmi leukocyty s t\u00edm, \u017ee kone\u010dn\u00fd roztok sp\u00e1lil a zbytky uva\u0159il a pova\u0159il v alkoholu.<\/p>\n\n\n\n

    Co vedlo Mieschera a Hoppe-Seylera k p\u0159esv\u011bd\u010den\u00ed, \u017ee prom\u00fdv\u00e1n\u00ed kyselinou zni\u010d\u00ed a odstran\u00ed pouze bun\u011b\u010dn\u00e9 st\u011bny a cytoplazmu a j\u00e1dro a jeho obsah ponech\u00e1 neporu\u0161en\u00e9 a dokonale zachovan\u00e9? Mnoh\u00e9 chemick\u00e9 l\u00e1tky maj\u00ed schopnost smr\u0161\u0165ovat bu\u0148ky, dehydratuj\u00ed bu\u0148ky, bylo toto br\u00e1no v \u00favahu p\u0159i pozorov\u00e1n\u00ed chemicky o\u0161et\u0159en\u00fdch leukocyt\u016f nebo jader?<\/p>\n\n\n\n

    Je t\u0159eba m\u00edt na pam\u011bti n\u011bco velmi d\u016fle\u017eit\u00e9ho: obsah bun\u011bk je pod mikroskopem viditeln\u00fd jen z\u0159\u00eddka, zejm\u00e9na je\u0161t\u011b v roce 1860; a pokud viditeln\u00e9 jsou, tak jen n\u011bkter\u00e9 slo\u017eky, nap\u0159. j\u00e1dro, mitochondrie. Miescherovo pozorov\u00e1n\u00ed po chemick\u00e9m o\u0161et\u0159en\u00ed mohou b\u00fdt ve skute\u010dnosti zmen\u0161en\u00e9 leukocyty.<\/p>\n\n\n\n

    Pokud se pod\u00edv\u00e1te na videa leukocyt\u016f pod mikroskopem, zjist\u00edte, \u017ee aktivn\u00ed a pohyblivou \u010d\u00e1st\u00ed t\u011bchto bun\u011bk je cytoplazma. Pohyb leukocyt\u016f vykon\u00e1vaj\u00ed l\u00e1tky v cytoplazm\u011b; j\u00e1dro z\u016fst\u00e1v\u00e1 neaktivn\u00ed, pouze m\u011bn\u00ed tvar na z\u00e1klad\u011b aktivity cytoplazmy. Zaj\u00edmalo by m\u011b, co vede v\u011bdce k p\u0159esv\u011bd\u010den\u00ed, \u017ee kl\u00ed\u010d k \u017eivotu a d\u011bdi\u010dnosti se nach\u00e1z\u00ed v n\u011b\u010dem neaktivn\u00edm, v n\u011b\u010dem tak pasivn\u00edm.<\/p>\n\n\n\n

    Definice skute\u010dn\u00e9 extrakce nebo izolace je “vyjmut\u00ed z\u00e1jmov\u00e9 \u010d\u00e1stice z ostatn\u00ed hmoty”. V Miescherov\u00fdch a Hoppe-Seylerov\u00fdch pokusech nedo\u0161lo k izolaci v \u017e\u00e1dn\u00e9m bod\u011b procesu, nejl\u00e9pe by jejich pokusy popsalo chemick\u00e9 prom\u00fdv\u00e1n\u00ed lidsk\u00fdch a ps\u00edch exkrement\u016f.<\/p>\n\n\n\n

    Miescherovy a Hoppe-Seylerovy v\u00fdzkumn\u00e9 pr\u00e1ce bohu\u017eel neobsahuj\u00ed \u017e\u00e1dn\u00e9 n\u00e1kresy izolovan\u00fdch bun\u011bk, konkr\u00e9tn\u011b obsahu, kter\u00fd pozorovali pod mikroskopem p\u0159ed a po jednotliv\u00fdch kroc\u00edch.     Miescher zjistil, \u017ee z\u00edskal novou l\u00e1tku, kterou by mohl b\u00fdt nuklein, na z\u00e1klad\u011b n\u00e1sleduj\u00edc\u00edch pozorov\u00e1n\u00ed:<\/p>\n\n\n\n

        V podstat\u011b dosp\u011bl k z\u00e1v\u011bru, \u017ee objevil novou l\u00e1tku na z\u00e1klad\u011b reakce z\u00edskan\u00e9ho roztoku na pou\u017eit\u00e9 chemik\u00e1lie a postupy, a nikoli na z\u00e1klad\u011b skute\u010dn\u00e9 izolace a pozorov\u00e1n\u00ed obsahu j\u00e1dra pod mikroskopem.<\/p>\n\n\n\n

    Kyselina fosfore\u010dn\u00e1 je bezbarv\u00e1 kapalina a dus\u00edk je plyn; oboj\u00ed je pom\u011brn\u011b nebezpe\u010dn\u00e9. N\u00e1lez t\u011bchto l\u00e1tek po chemick\u00e9 alkalizaci, okyselen\u00ed, vyva\u0159en\u00ed a sp\u00e1len\u00ed v\u0161eho, co zbylo, vypov\u00edd\u00e1 jen velmi m\u00e1lo o molekul\u00e1rn\u00ed struktu\u0159e tk\u00e1n\u011b, bun\u011bk, j\u00e1dra a nukleinu, zejm\u00e9na v jejich \u017eiv\u00e9m stavu. Obecn\u011b n\u00e1lez \u010dehokoli po zm\u00edn\u011bn\u00fdch chemick\u00fdch l\u00e1tk\u00e1ch a kroc\u00edch vypov\u00edd\u00e1 jen velmi m\u00e1lo krom\u011b toho, \u017ee odst\u0159e\u010fov\u00e1n\u00edm, va\u0159en\u00edm, zah\u0159\u00edv\u00e1n\u00edm a spalov\u00e1n\u00edm chemicky o\u0161et\u0159en\u00e9 tk\u00e1n\u011b vznikaj\u00ed nebezpe\u010dn\u00e9 l\u00e1tky.<\/p>\n\n\n\n

    Opakov\u00e1n\u00ed postup\u016f a dosa\u017een\u00ed stejn\u00fdch v\u00fdsledk\u016f, tedy Hoppeho-Seylerovy pokusy, neprokazuj\u00ed, \u017ee byla nalezena nov\u00e1 l\u00e1tka. Opakov\u00e1n\u00ed stanov\u00ed, \u017ee p\u0159i zpracov\u00e1n\u00ed podobn\u00e9 l\u00e1tky podobn\u00fdmi chemick\u00fdmi l\u00e1tkami a p\u0159i pou\u017eit\u00ed podobn\u00fdch postup\u016f vzniknou stejn\u00e9 vedlej\u0161\u00ed produkty.<\/p>\n\n\n\n

    Zat\u00edmco Miescher z\u00edskal leukocyty z hnisu na obvazech, Hoppe-Seyler se z nezn\u00e1m\u00e9ho d\u016fvodu rozhodl z\u00edskat leukocyty ze ps\u016f tak, \u017ee jim do b\u0159icha vlo\u017eil \u010do\u010dky a pozd\u011bji tyto psy usmrtil za \u00fa\u010delem extrakce a vy\u0161et\u0159en\u00ed. Vzhledem k tomu, \u017ee existuje neinvazivn\u00ed a \u017eivot ni\u010d\u00edc\u00ed zp\u016fsob, jak z\u00edskat leukocyty, jak\u00fd je d\u016fvod prov\u00e1d\u011bt tak drsn\u00e9 pokusy? Extrakce r\u016fzn\u00fdch l\u00e1tek se prov\u00e1d\u011bla va\u0159en\u00edm \u010d\u00e1st\u00ed t\u011bl ps\u016f v alkoholu a\/nebo v \u017e\u00edrav\u00e9 sod\u011b a n\u00e1sledn\u00fdm okyselen\u00edm sm\u011bsi kyselinou octovou. K testov\u00e1n\u00ed p\u0159\u00edpadn\u00e9 reakce se p\u0159id\u00e1val tak\u00e9 vitriol, roztok sody a oxid bizmutit\u00fd. Hoppe-Seyler\u016fv pokus na psech mi p\u0159ipomn\u011bl pokusy Louise Pasteura, kter\u00fd v roce 1885 tak\u00e9 mu\u010dil psy, aby vyvinul vakc\u00ednu proti vzteklin\u011b.<\/p>\n\n\n\n
